الأستروجين
الأستروجين هو هرمون يتم انتاجه أساسا في المبيض والقليل منه يأتي من بعض الأنسجة الأخرى مثل الدهون.
ثلاثة أنواع شهيرة منه هي الأستروجين الأولي أسترون estrone ويوجد في السيدات بعد انقطاع الدورة وفي الأطفال والثنائي estradiol الأكثر انتشارا وهو ينتج من بويضات المبيض مع تغيرات الدورة الشهرية أما الثلاثي estriolفهو موجود في الحوامل من نشاط المشيمة.
مصادر الأستوجين الدوائي إما طبيعي وتأتي من النبات كالأستراديول أو من الحيوان كالبريمارين من أنثى الحصان
أو مصنع مثل ethinylestradiol and estradiol valerate.
لكي يعمل الأستروجين لابد له من مستقبلات خاصة في جدار الخلية ليعبر للداخل وفي النواة نفسها ليعطي تأثيره. يوجد نوعان من المستقبلات في النواة مستقبل ألفا ومستقبل بيتا nuclear estrogen receptors (ERα and ERβ). .
يعض أنواع الأستروجين النباتي والمصنع لها تأثير معين على مستقبلات ألفا أو بيتا في موضع وعكس هذا التأثير في مكان آخر اسم هذه المجموعة مغيرات مسقبلات الأستروجبن (SERMs) ومن أمثلة ذلك كلوميفين و تاموكسيفين ورالوكسيفين وتيبولون.
ولأن هذه تعطل المستقبلات أو تنشطها في أماكن معينة فهي تستعمل لأهداف محددة كتنشيط التبويض أو تثبيط سرطان الثدي ووظائف أخرى كثيرة.

الدور الأنثوي بإحداث البلوغ والتغيرات الجسمية
الدور الخصوبي فهو ضروري للانجاب
المحافظة على صحة الجدار الداخلي للمهبل والافرازات الصحية.
المحافظة على الأوعية الدموية
سلامة الثدي والعظام والجلد وميزان التجلط
التأثير النفسي وتأثير على وظائف الجهاز العصبي
يستعمل هذا الهرمون في منع الحمل وعلاج المتاعب المصاحبة لانقطاع الدورة وعلاج هشاشة العظام

Estrogens
Estrogens are produced by the o***y and other tissues – notably adipose tissue. The types of estrogens are 17β estradiol, estrone and estriol and have steroidal biochemical structure which allows them to pass through the phospholipid membrane of the cell and reach the nucleus.
All estrogenic effects depend on whether a cell has estrogen receptors or not. Without specific receptors, cells do not respond to the presence of estrogens. Receptors are on the cell membrane and/or in the nucleus, with the nuclear receptors being the most important. There are 2 types of nuclear estrogen receptors (ERα and ERβ). The 2 receptors are of different genetic origins (located on different chromosomes) but have similar functions in bodily tissues. The tissues in which they are found are: endometrium, breast, o***y, kidney, brain (notably the hypothalamus), bone, heart, lungs, intestinal mucosa, prostate and endothelial cells.
These receptors are present or absent in breast cancer lesions which are described as being ER positive or ER negative and their presence (or not) dictates regimens of therapy and prognosis.
Estrogens come with various actions or potency depending on how they bind onto the receptor sites. The endogenous estrogens, mainly estradiol and some animal produced hormones (for example conjugated equine estrogens – Premarin©) have positive or agonistic effects. Synthetically produced estrogens such as ethinylestradiol and estradiol valerate are also agonists and are used in medications such as oral contraceptives.
Some estrogens have mixed effects, being promotive (agonists) or inhibitory (antagonists) on different tissues. There are some plant estrogens and synthetic agents that selectively modulate the receptors. They are called Selective Estrogen Receptor Modulators (SERMs) and examples are clomiphene, tamoxifen, raloxifene and tibolone.
Because they selectively enhance or block estrogenic activity SERMs can encourage ovulation, suppress breast cancer as adjuvant therapy, re**rd osteoporosis, reverse vaginal atrophy and possibly provide endometrial protection when combined with menopausal hormone therapy. Much research continues to be conducted to find niche uses for SERMs.
The functions of estrogens include:
• The development and regulation of the female reproductive system, the initiation of puberty and the proliferation of the endometrium
• Maintenance of secondary s*x characteristics
• Vaginal lubrication and thickening
• Maintenance of blood vessel endothelial function
• Skin elasticity, collagen content and thickness
• Breast and bone integrity
• Coagulation and modulatory central nervous system function
• Use in oral contraceptives, menopausal hormone therapy, osteoporosis and hypogonadal states
• Anti-estrogen therapies (such as tamoxifen and aromatase inhibitors) are used to inhibit breast cancer growth.
