Codeine dysregulates ribosome biogenesis in Escherichia coli with DNA double-strand breaks to chart path to new classes of antibiotics

Abstract
Aim: A bacterial genetics-guided approach was utilized for the discovery of new compounds affecting bacterial genome stability. Materials & methods: Fungal extracts and fractions were tested for genome instability-mediated antibacterial activity. Interaction assays and RT-qPCR were used to identify compounds that boost the activity of sub-minimum inhibitory concentration streptomycin and obtain insights on the molecular mechanisms of the primary hit compound, respectively. Results: Several extracts and fractions caused bacterial genome instability. Codeine, in synergy with streptomycin, regulates double-strand break (DSB) repair and causes bacterial ribosome dysfunction in the absence of DSBs, and dysregulation of ribosome biogenesis in a DSB-dependent manner. Conclusion: This study demonstrates a potential viable strategy that we are exploring for the discovery of new chemical entities with activities against Escherichia coli and other bacterial pathogens.

Imagination is more important than knowledge: Inventing a TLC sprayer

We live by this idea daily in our small and struggling laboratory for chemical systems biology. Struggling yet bold with our ideas and ambitions.

This is a story of the power improvisation within low resources settings. For months now we have been trying to procure spraying device for thin layer chromatography. The device we wanted is a bit expensive and the procurement is taking forever.

So got to work to find a local replacement
which will work fine. All the options we tried fall apart after a few rounds of use due to the strong acid in the staining reagent.

So we went back to town and found this item that looks like it's made with durable plastic. It's used for spreading insecticides and cleaning solutions.

We could vouch for the spraying mechanism to survive so this quick thinking brother decided to separate the problem into two and limit the exposure of strong acid only to the nozzle side and spare the air pump.

He tried a long test tube first and then we thought of this little flask. At least it reduces the number of times we need to refill.

We tested it and worked beautifully. The nozzle is tunable which allows us to obtain super fine spraying. And it is very durable too. We have used it intensely for two months now and it's going strong.

Again our quick thinking brother decided to be over cautious. And he rinses the device with deionized water each time after use in order not to give the strong acid the chance to drift into the air pump side. The soft plastics that allows the piston to deliver the pressure will not survive.

It looks like with this strategy we can going to have this improvised device around for a long time. And costed us just about 7 USD instead 190 USD that would have taken forever to arrive.

Feel free and set one for your Laboratory if you don't have a good sprayer at hand. This is better than pouring the reagent over the plate or even dipping into a tank of reagent.
