In a major leap for cancer treatment, scientists have found a way to create cancer-killing immune cells inside the body—using a vaccine-like shot made with mRNA. Until now, CAR T-cell therapy involved a complex and expensive process: extracting a patient’s T cells, engineering them in a lab to target cancer, and reinfusing them back into the body. While this approach has saved lives, it takes weeks and involves harsh chemo to prepare the body. The new method could change everything.

Instead of removing cells, researchers used a modified mRNA shot—similar to the ones used for COVID-19—to deliver cancer-fighting instructions directly into the bloodstream. Once inside, the instructions reprogram normal T cells into CAR T cells within hours. In trials on mice, rats, and monkeys, this approach successfully destroyed cancer cells, lowered dangerous B cell levels, and lasted for at least a month with minimal side effects.

Unlike DNA-based therapies that risk altering a cell’s genome, mRNA is temporary and doesn’t damage DNA, making it potentially safer. The researchers designed special lipid nanoparticles that skipped the liver (a common barrier) and targeted immune cells directly. These fat-based shuttles carried mRNA that taught the T cells how to recognize and kill specific cancer cells.

Even more promising, this technique may help reset the immune system in autoimmune conditions like lupus, where B cells attack the body. A Phase 1 human trial is being planned, with hopes to expand this method to other diseases.
