We seek to improve our patients´ quality of life, decrease their disability and reduce the impact of degenerative joint disease on healthcare systems worldwide by preserving joints and maintaining function. A recent study establishes a novel ex vivo bovine explant model using skeletally immature developing joints to investigate the formation of the zone of calcified cartilage. The authors demonstrate that cartilage calcification is a highly zonal, cell-driven process: the deep cartilage layer actively promotes calcification, whhereas the superficial layer inhibits it through secreted factors. Using osteochondral and split-layer cartilage cultures, they show that viable chondrocytes in the top layer suppress calcification in the bottom layer, whereas loss of viability removes this inhibitory control. Gene expression analyses reveal a clear spatial signature, with calcification inhibitors (PTHLH, MGP, FGF18) enriched in the superficial zone and hypertrophic/calcification markers (COL10A1, ALPL, IHH) dominant in the deep zone. This model provides a physiologically relevant platform to study calcified cartilage formation and opens new avenues for understanding osteochondral regeneration and pathological calcification in osteoarthritis.
