A groundbreaking therapy, based on inhibiting the USAG‑1 protein, is now undergoing human clinical trials in Japan to test whether it can regenerate missing teeth—potentially even a third set—by reactivating dormant tooth buds. This is real, ongoing research.

What's happening now:

Human trials began around September or October 2024. These studies involve 30 healthy adult males (ages 30–64), each missing at least one tooth, and aim to assess the safety and dosing of the drug when administered intravenously.

The drug works by blocking USAG‑1, a protein that suppresses tooth development; preclinical successes include regrowing teeth in mice and ferrets.

If adult trials prove safe and effective, the plan is to expand testing to children (ages 2–6 or 2–7) with congenital tooth agenesis.

Experts project public availability by around 2030, contingent on trial success and regulatory approval.

Broader context:

This therapy could potentially offer an alternative to dentures and implants, especially for those with congenital tooth loss or injury-related tooth loss.

Meanwhile, other global efforts in tooth regeneration are also underway—such as lab-grown teeth using scaffolds (Tufts University), enamel-forming proteins (University of Washington), and fully integrated lab-grown teeth (King’s College London)—suggesting a broader push toward regenerative dentistry.

According to Wikipedia, the first clinical trial for tooth regeneration in Japan started around 2023 using a medicine aimed at stimulating tooth regrowth via USAG‑1 inhibition.
