Under methylation can cause lower levels of serotonin, dopamine, and noradrenaline in the system. This is typically due to a faster reuptake of these specific neurotransmitters at the nerve synapse (the junction where nerves meet) within the brain.

Phase 1 estrogen detoxification pathways can lead to the short-lived yet metabolically dynamic 2-OH, 4-OH, and 16-
OH estrogen metabolites.
These metabolites have varying effects on the body before being detoxified - understanding their differences can provide insight into symptoms, health conditions, and potential health risks related to estrogen metabolism.
2-OH:
- This pathway is considered the “preferred” pathway because they are generally more stable, cause the least harm, and may even have protective effects against cancer growth.
4-ОН:
- These are deemed the most “carcinogenic” or cancer-prompting of metabolites because they can turn into highly reactive molecules that damage our DNA if not quickly or adequately processed into a stable form.

16-ОН:
- These are the more “proliferative” metabolites because they promote cell growth.
- They bind strongly to estrogen receptors relative to the 2-OH metabolites, which may be helpful with bone health; however, they may be harmful for breast cancer, uterine fibroid development, heavy menstrual bleeding, and endometriosis due to their proliferative nature.

The Role of Genes in Methylation

Emphasising the accuracy of test results analysis is of paramount. An excellent example of this is the examination of oxidative damage in tissues and cells, which involves a redox reaction/cycle. We not only test specific genes associated with this process but also test a combination of glutathione markers and 8OHDG to assess the extent of damage to our DNA. While testing only glutathione provides a momentary snapshot within the normal range, incorporating the 8OHDG marker enables us to gain a comprehensive understanding of the oxidative stress inflicted on our DNA. Subsequently, we investigate the root causes of this damage. The inclusion of 8OHDG in our routine baseline tests is increasingly prevalent as both our practitioners and clients want to continuously strive to minimise its levels and effects. This can be tested in isolation but is also included in the DUTCH Complete test.

Gut dysbiosis is an imbalance or disruption in the normal composition of the gut microbiota, which is the community of microorganisms residing in the gastrointestinal tract. This imbalance can lead to various health issues, including inflammation, which can subsequently contribute to high 5-alpha-reductase (5α-reductase) enzyme activity, elevated dihydrotestosterone (DHT) levels, and the development of acne and baldness in males.

1.Gut dysbiosis and inflammation: The gut microbiota plays a crucial role in maintaining a balanced immune system and promoting overall health. When the gut microbiota becomes imbalanced, with an overgrowth of harmful bacteria or a decrease in beneficial bacteria, it can trigger an inflammatory response in the gut. This inflammation can result in increased intestinal permeability, also known as "leaky gut," allowing toxins, bacterial byproducts, and other molecules to leak into the bloodstream. This leakage can activate the immune system and lead to systemic inflammation throughout the body.

2. Increased 5α-reductase enzyme activity: Within the body, the enzyme 5-alpha-reductase converts testosterone, a male s*x hormone, into dihydrotestosterone (DHT). DHT is a more potent androgen and plays a role in various physiological processes, including the development of male s*xual characteristics. However, an imbalance in gut microbiota and subsequent inflammation can potentially increase the activity of 5α-reductase enzyme, leading to higher levels of DHT.

3. Elevated DHT and its impact on acne: Elevated levels of DHT can have effects on the skin and hair follicles. In the case of acne, high DHT levels can stimulate the sebaceous glands in the skin to produce more sebum, an oily substance. Excessive sebum production, combined with other factors like clogged pores and the presence of bacteria, can contribute to the development of acne.

What are SNPs?

SNPs, or Single Nucleotide Polymorphisms, are variations at a single position in a DNA sequence among individuals. They are the most common type of genetic variation in humans.

1. Homozygous: This term is used when an individual has two identical forms of a particular gene, one inherited from each parent. In the context of SNPs, a person is homozygous for a SNP if the same variation is present on both copies of the DNA sequence. Depending on the specific gene, homozygous can affect the function of an enzyme by up to 70%

2. Heterozygous: This term is used when an individual has two different forms of a particular gene, one inherited from each parent. In the context of SNPs, a person is heterozygous for a SNP if different variations are present on the two copies of the DNA sequence. Depending on the specific gene, homozygous can affect the function of an enzyme by up to 30%

In simple terms, if you have two of the same version of a gene or SNP, you're homozygous. If you have two different versions, you're heterozygous.

What is it??????

Higher visceral fat (belly fat) causes an increase in a specific enzyme known as aromatase. This enzyme converts our testosterone into oestrogen. When oestrogen levels begin to rise in men a number of symptoms arise:

- Lower muscle mass
- Lower s*x drive/Libido
- Increase breast tissue
- Depression

It is equally important to ensure optimal methylation as it is to avoid over-treatment resulting from purely genetic SNP testing, a frequent occurrence in our clinics. While understanding your genetic SNPs is crucial, it should always be correlated with your methylation status. Many clients arrive at our clinics with MTHFR gene SNPs, taking very high doses of methyl folate supplements, yet they have no idea of their methylation status. Oestrogen metabolites and bone health in menopause: Adequate levels of 16OH oestrogen, post-menopause are important for bone health. Oestrogen plays a crucial role in maintaining bone density and strength in both men and women. During menopause, there is a natural decline in oestrogen production, which can lead to accelerated bone loss and an increased risk of osteoporosis. Oestrogen affects bone health in several ways. It helps to regulate the balance between bone formation and resorption, the process by which old bone is broken down and replaced with new bone tissue. Oestrogen also helps to inhibit the activity of osteoclasts, cells that are responsible for breaking down bone. When oestrogen levels decrease, as in post-menopausal women, the balance shifts in favor of bone resorption, leading to a net loss of bone density. Studies have suggested that 16OH oestrogen may have a protective effect on bone by promoting bone formation and inhibiting bone resorption. It is believed to have a stronger bone-protective effect compared to other oestrogen metabolites including both 2OH & 4 OH oestrogens. Green Tea 🍵 Super healthy yeh?

Not with an AA COMT SNP

Our COMT gene is responsible for metabolising oestrogen, dopamine, adrenaline & noradrenaline but also catecholamine containing foods. One rich source of these foods is green tea. Although a potent antioxidant for people with slower COMT function or AA genetic snps, higher green tea consumption can cause anxiety, panic and also oestrogen dominant symptoms (migraine, breast tenderness, heavy bleed, cramping, PMT etc). Although genes are not our destiny, knowing their effects can allow us to individualise not alone nutraceuticals but also diet itself.

Serotonin……..

is a neurotransmitter that is synthesised, stored, and released by neurons in the brain, where it plays a role in mood, cognition, reward, learning, memory, and other functions. However, it's important to note that the majority of the body's serotonin (about 90%) is produced in the gut, where it primarily helps regulate intestinal movements.

Serotonin cannot cross the blood-brain barrier, which means that serotonin produced in the gut cannot directly reach the brain. Instead, the serotonin produced in the gut performs local actions in the gut and communicates with the brain indirectly through various other mechanisms.

For example, gut serotonin can stimulate the vagus nerve, a major neural pathway between the gut and the brain. This stimulation sends a signal to the brain and can influence mood and behavior. Furthermore, gut bacteria can also influence the production of serotonin in the gut, which adds another layer of complexity to the gut-brain axis.

It's also worth noting that certain precursor molecules and cofactors required for serotonin synthesis (like the amino acid tryptophan and the cofactor vitamin B6) can cross the blood-brain barrier and influence serotonin production in the brain. Therefore, dietary and gut microbial influences on the availability of these precursors and cofactors can indirectly affect brain serotonin synthesis.

What is GI Map?

The Gastrointestinal Microbial Assay Plus (GI-MAP) is an innovative clinical tool that measures gastrointestinal microbiota DNA from a single stool sample with state of the art, quantitative polymerase chain reaction (qPCR) technology.

The GI-MAP was designed to detect microbes that may be disturbing normal microbial balance or contributing to illness as well as indicators of digestion, absorption, inflammation, and immune function.
