Rheumatoid Arthritis (RA) is an autoimmune disease. It is perhaps the most common inflammatory arthritis, affecting 0.5-1% of the general population worldwide. It is a systemic disease which not only causes joint problems but also can affect other organs. The precise cause for this disease is uncertain however environmental and genetic influences are clearly factors. These multiple factors interact to create and perpetuate RA. The initiation of RA begins years before the onset of clinical symptoms. The onset involves certain specific genes that can lead to production of Autoantibodies and lead to autoreactivity. Genes play a key role in RA susceptibility and severity. Newly defined genetic associations in more than 100 genes including polymorphisms in PTN22, PAD14, and genes involved in immune function suggest that the associations in RA are complex and involve many pathways. The microbiome and environmental stress at mucosal surfaces contribute to the etiology of RA. This disease predominates in women.
