📢 📢 Mazumdar Shaw Medical Foundation News & Updates 📢 📢

1. Anti-CXCR4 inhibitor in reversing resistance to chemoprevention and chemotherapy (Patent No.: 449011)
Patentee: Mazumdar Shaw Medical Foundation, Bangalore
Name of Inventor(s): Dr Moni Abraham Kuriakose, Dr. Amritha Suresh, Ms. Gangotri Siddappa, Dr. Safeena Kulsum
This is a method patent for reversing resistance to chemopreventive and chemotherapeutic drugs. It is an in vitro study, wherein the drug resistance is shown to increase on interaction of epithelial cells with fibroblast cells. Subsequent inhibition of this stromal interaction by blocking the activity of SDF1/CXCR4 pathway using anti-CXCR4 has indicated sensitization of epithelial cells to drugs and hence, killing the dysplastic/cancer cells. Further, development of in-house anti-CXCR4 molecules is being carried out currently.

2. A pharmaceutical formulation comprising curcumin and metformin for head and neck cancer (Patent No.: 462241)
Patentee: Mazumdar Shaw Medical Foundation, Bangalore
Name of Inventor(s): Dr Moni Abraham Kuriakose, Dr. Amritha Suresh, Ms. Gangotri Siddappa, Dr. Safeena Kulsum
This is a pharmaceutical formulation patent that comprises of use of both curcumin and metformin drugs together for chemoprevention of head and neck cancer. The efficacy of both the drugs individually are shown in various cancers including that of head and neck cancer. Here, in our study we have shown that the combination is more efficient as compared to their individual efficacy. A Phase IIb/III trial (Study title: Phase IIb/III study to determine the efficacy of Curcumin and Metformin to reduce the incidence of second primary tumors in aero-digestive tract in patients with history of head and neck squamous cell carcinoma) is also ongoing at Mazumdar Shaw Medical Centre, Narayana Health City, Bangalore for the same drug combination.

CD44-SNA1 integrated cytopathology for delineation of high grade dysplastic and neoplastic oral lesions September 2023 PLoS ONE DOI: 10.1371/journal.pone.0291972

Low patient compliance for biopsies of oral lesions underscores the need for less invasive diagnosis. The primary objective of this study was to ascertain the significance of biomarkers to improve cytology. Towards this effect, a comprehensive systematic review and meta-analysis identified a panel of markers (n: 10) delineating dysplastic oral lesions. Subsequent patient-based validation in tissues (n: 131) identified a four-marker panel, CD44, Cyclin D1, SNA-1, and MAA, with the best sensitivity (>75%; AUC>0.75), while validation by cytology in an independent cohort (n: 133) showed that SNA-1 and CD44 significantly delineate moderate-severe dysplasia (High-grade lesions: HGD) and cancer with high sensitivity (>83%). Multiplex validation in another cohort (n: 138), integrated with a machine learning model incorporating clinical parameters, further improved the sensitivity and specificity (>88%). Image automation using a single marker dataset (SNA-1), wherein a model was developed from morphological and expression features also gave a high sensitivity (86%). Our study indicated that the use of a two-biomarker panel (CD44/SNA-1) integrated with clinical parameters, when multiplexed or integrated with automated image analysis provided efficient risk stratification of oral lesions, indicating the significance of biomarker-integrated cytopathology in the development of a Point-of-care assay.
