What is the meaning of Primary Wavelength & Secondary wavelength

➡️ λ₁ is the main wavelength used to measure analyte absorbance.

➡️ λ₂ is reference wavelength used to correct interference (turbidity, sample colour etc.)

➡️ E.g. Bilirubin T&D

Primary Wavelength (λ₁):
546 nm - Measures the true absorbance of the purple azobilirubin complex.

Secondary Wavelength (λ₂):
600 nm - Corrects background interference (due to turbidity or sample colour).

What is the meaning of λmax & λoptima
λmax:
➡️Wavelength at which the reaction shows maximum absorbance.
➡️ For glucose: λmax - 505 nm but also prone to interference of other molecules e.g. Hemoglobin, Bilirubin etc.
λoptima:
➡️ Optimum wavelength selected by the analyzer for a specific test.
➡️ For glucose λoptima is 540 – 546 nm. At this wavelength no interference of Hemoglobin, Bilirubin etc.

What is the meaning of Interval Time
➡️ Interval Time is the time gap between two consecutive absorbance (signal) readings taken by the Analyzer during the measurement phase (Read Time).
➡️ It defines how frequently the Analyzer looks at the reaction progress.

Cholesterol Test
➡️ All cholesterol esters present in plasma are hydrolysed into free cholesterol and fatty acids by cholesterol esterase.

➡️ In the presence of oxygen, free cholesterol is then oxidized by cholesterol oxidase to cholestenone and H2O2. H2O2 then reacts with chromogen in the presence of peroxidase to form a red dye.

➡️ The intensity of color formed is proportional to the cholesterol concentration and can be measured photometrically at 546 nm.

➡️ Cholesterol is estimated by End-Point mode.
