A groundbreaking new drug that could help people regrow missing teeth has entered clinical trials, marking a major milestone in dental medicine.

Developed by a team led by Katsu Takahashi, head of oral surgery at the Medical Research Institute Kitano Hospital in Osaka, the drug is designed to activate dormant tooth buds—remnants of a potential third set of teeth hidden beneath the gums.

While humans typically grow only two sets of teeth, these hidden buds might be reawakened to generate new ones.

The clinical trials, launched in October at Kyoto University Hospital, involve giving the experimental drug to adult volunteers.

The treatment targets a protein called USAG-1, which appears to suppress natural tooth regeneration.

Animal studies showed that blocking this protein enabled the growth of new teeth—results researchers hope to replicate in humans.

If successful, the drug could offer a less invasive and more affordable alternative to traditional dental prosthetics like implants and dentures.

The initial focus is on children with a rare condition that causes them to be born with fewer than six permanent teeth—a disorder that affects about 0.1% of the population and can severely impair chewing.

The researchers aim to make the drug publicly available by 2030.

Although the trial’s primary goal is to ensure safety, Takahashi is optimistic that it may also trigger tooth regrowth, potentially revolutionizing dental care.

Researchers at King’s College London and Imperial College London have bioengineered early-stage human teeth in a lab using a hydrogel scaffold.

This gel mimics the environment of early tooth development and allows human dental pulp stem cells to grow into structured tissues resembling real teeth.

The hydrogel enables cell signaling and tissue organization critical for forming enamel and dentin-like structures.

This discovery may pave the way for regenerative dentistry, replacing synthetic fillings, crowns, or implants with naturally grown teeth.

“Scientists Are Testing A Drug That Could Regrow Human Teeth - Just Like Sharks Do, Might Replace Implants”

Japanese scientists have begun human trials of a new drug designed to regrow human teeth, offering a potential alternative to traditional implants and dentures. The drug, TRG-035, works by inhibiting the USAG-1 protein, which suppresses tooth development.

In animal trials, it successfully activated dormant tooth buds and led to new tooth growth in mice and ferrets. The initial human trial, underway at Kyoto University Hospital, involves 30 adults aged 30 to 64 who are missing at least one tooth. This first phase will assess the drug's safety before moving to pediatric trials targeting children with congenital tooth agenesis, a condition where permanent teeth never form.

If successful, the researchers hope to launch the treatment commercially by 2030, positioning it as a revolutionary biological solution to tooth loss and a potential replacement for artificial implants.
