Quantitative information at the proteome level aids in revealing the roles of proteins in biological, physiological and pathological processes. The most widely used strategies in mass spectrometry-based proteome-wide quantitative approaches, other than label-free quantification, are based on stable isotope labeling, which allows us to analyse multiple samples simultaneously in a single LC-MS run.

To reduce the complexity of MS2 spectra of IPTL methods and to further improve multiplexing capacity, researcher of the consortium propose a novel labeling approach that relies on a collision-induced dissociation (CID)-cleavable isobaric Ac-IPG tag. In this method, quantitative information is concealed in the Ac-IPG tag, which is located on the ε-amino group of the C-terminal Lys residue of LysC peptides and revealed upon tandem MS in the form of y-ion clusters containing different numbers of .

Quantifying proteins based on peptide-coupled reporter ions is a multiplexed quantitative strategy in proteomics that alleviates the problem of ratio distortion caused by peptide cofragmentation, as commonly observed in other reporter-ion-based approaches, such as TMT and iTRAQ. Data-independent acquisition (DIA) is an attractive alternative to data-dependent acquisition (DDA) due to its better reproducibility.

To establish a new peptide-coupled reporter-ion-based tag that avoids ratio distortion in DDA and improves the throughput of DIA-based methods without sacrificing the data acquisition rate, researcher of the consortium propose a novel isobaric Ac-AG tag, which conceals quantitative information in isobarically labeled peptides and reveals it upon tandem MS in the form of peptide-coupled reporter ions. Being the most common and the most aggressive subtype among renal cancers, clear cell renal cell carcinoma (ccRCC) accounts for most RCC fatal outcomes. Such outcomes probably arise from significant intra-tumor and inter-tumor genetic diversity of ccRCC, revealed in recent genomic studies. Indeed, among genetic factors that contribute to RCC risk, the most investigated is mutation of VHL gene, underlying the abnormal accumulation of hypoxia-inducible factor α (HIFα) proteins in normoxia.

(RCC) represents a group of histologically similar neoplasms with significant intratumor and intertumor genetic heterogeneity. Recognized factors for RCC development include smoking, hypertension, obesity, as well as von Hippel–Lindau (VHL) disease. For more than a decade, it has been suggested that a complex interplay between and chronic inflammation represents one of the underlying mechanisms of gradual cardiac depression in (HF). Oxidative stress in HF is believed to be a consequence of increased circulating neurohormones and hemodynamic disorder, as well as inflammation and decreased oxygen delivery.

Having all that in mind, Researcher of the consortium conducted a pilot case-control study consisting of patients with HF due to coronary artery disease ( ) or idiopathic dilated cardiomyopathy ( ) in order to compare the distribution of common GST genotypes and the differential risk effect between these two entities.
