1. Objective:

The objective of the blood grouping test was to determine the ABO and Rh (D) blood group of a person by observing agglutination reactions.
---

2. Principle:

The test was based on antigen-antibody reactions. When red blood cells (RBCs) possessing specific antigens (A, B, or RhD) were mixed with corresponding antisera (Anti-A, Anti-B, and Anti-D), visible agglutination occurred, indicating the presence of that antigen.

3. Materials:

Clean glass slide or blood grouping card

Antisera (Anti-A, Anti-B, and Anti-D)

Sterile lancet

Cotton swab with alcohol

Dropper or applicator sticks

Blood sample (capillary or venous)

4. Procedure (Microscopic not required; this is a macroscopic test):

1. The glass slide or card was labeled with three sections: Anti-A, Anti-B, and Anti-D.

2. One drop of each antisera was placed on the respective sections.

3. A finger was pricked using a sterile lancet, and a drop of blood was placed in each section.

4. Each mixture was stirred using a separate stick to avoid cross-contamination.

5. The slide was gently rocked for about 1 minute, and agglutination (clumping) was observed with the naked eye.

Agglutination with Anti-B only: Blood group B

Agglutination with both Anti-A and Anti-B: Blood group AB
