In order to separate PRP from blood, it is important not only which kit you put the blood into, but also which centrifuge you set and how you centrifuge it.
Normally, when extracting PRP from blood using a centrifuge, a low-speed centrifuge is usually used.
Kits that can separate PRP usually provide instructions on how to set the "rpm" and "operating time" of the centrifuge.
However, even if the "rpm" is the same, the "rcf value" may differ between centrifuges, so the "rcf value" during the centrifugation period must also be checked. And the centrifuge has a brake level until the centrifuge stops after the operating time is completed. b0 has the longest break time and b8 has the shortest break time so it stops immediately. Therefore, it is necessary to check how the b (brake) of the centrifuge is set. In particular, if you centrifuge blood for too long, RBCs may break or cells in the blood may be destroyed, so be sure to check the operation time.

Precautions when drawing blood to extract PRP
1. Use a 21-23 gauge needle.
2. If the needle is thick, pain may occur when collecting blood, and if the needle is too thin, RBCs may be destroyed and hemolysis may occur.
3. If RBCs are destroyed, PRP cannot be extracted from blood.
4. Remember that in order to extract PRP from collected blood using the PRS Kit, 90% of blood and 10% of anticoagulant must be mixed into the PRS Kit.
When the total amount is 30cc, 27cc of blood and 3cc of anticoagulant must be added, and when the total amount is 25cc, 22.5cc of blood and 2.5cc of anticoagulant must be mixed in the PRS Kit.

What anticoagulant is best to use when extracting PRP from blood?
It is best to use anticoagulants containing citrate.
Because the blood is separated through a centrifuge immediately after blood collection, there is no need to use anticoagulants that prevent the blood from coagulating for a long time.
Therefore, it is best to use Acid Citrate Dextrose (ACD) liquid, but Citrate Phosphate Dextrose (CPD) can also be used. However, heparin is not recommended.

The PRS Bio Kit is a kit that safely and easily extracts PRP from 25 cc (maximum 30 cc) of blood containing anticoagulant. The anticoagulant used when separating PRP should account for 10% of the total volume. In other words, when separating 25 cc, 22.5 cc of blood and 2.5 cc of anticoagulant must be mixed. This 7-minute video is a bit long, but it will be very helpful to those who want to know how to extract PRP from blood.
