Dr. Anne Beaven of UNC Lineberger leads Alliance A052101, a clinical trial designed to answer an important question for older adults with mantle cell lymphoma (MCL):
➡️ Can patients who achieve remission safely receive fewer treatments of the targeted therapy zanubrutinib—without compromising outcomes?
Zanubrutinib, a Bruton tyrosine kinase (BTK) inhibitor, has shown strong effectiveness in treating MCL. However, long-term continuous treatment can bring side effects, financial toxicity, and quality of life challenges, particularly for older adults. This study explores whether a reduced duration treatment approach could maintain disease control while lowering treatment burden.
Why this trial matters:

🔹 May reduce cumulative toxicity and improve daily quality of life
🔹 Supports a more individualized, remission guided treatment strategy

For patients with early-stage triple-negative breast cancer who achieve a pathologic complete response after chemotherapy and pembrolizumab, what’s next? Should treatment continue—or is monitoring enough?
The Alliance A012103 trial, led by Dr. Sara Tolaney at Dana-Farber, and is a phase III study designed to compare:
✅ Continuing pembrolizumab after surgery
✅ Monitoring only (“watch and wait”)
Why does this matter? If monitoring proves just as effective, patients could avoid unnecessary treatment, reduce side effects, and improve quality of life—while still maintaining excellent outcomes.
This research could reshape how we approach care for TNBC patients who respond well to initial therapy.

The B LuRE trial (A021901) study is now open for enrollment through the Alliance for Clinical Trials in Oncology!
Led by Thomas Hope, MD of UCSF and Suki Padda, MD of Fox Chase Cancer Center, this important trial is evaluating a new treatment approach for patients with bronchial neuroendocrine tumors.
🔬 What the study is testing:
B LuRE is comparing a radioactive drug (a form of targeted radiopharmaceutical therapy) with standard chemotherapy to see which option offers better outcomes for people living with bronchial neuroendocrine tumors.
This trial has the potential to advance care for a rare and often difficult to treat disease by exploring therapies that can more precisely target cancer cells while sparing healthy tissue.

Led by Adrienne Waks, MD, of the Dana Farber Cancer Institute, A012303 (ShortStop HER2), is Phase III clinical trial now open for enrollment through the Alliance for Clinical Trials in Oncology.
👉 This study asks if people with early stage HER2 positive breast cancer who have no cancer left (pCR) after neoadjuvant chemotherapy + HER2 targeted therapy, can safely have less treatment after surgery?
Many patients who achieve a pathologic complete response still receive additional therapy after surgery, even though we know pCR is associated with excellent outcomes. ShortStop HER2 aims to determine whether a shorter, less intensive approach can maintain effectiveness while reducing treatment burden, side effects, and cost.
If successful, this study could be practice changing by offering patients:
• A more personalized treatment pathway
• Lower toxicity
• Improved quality of life
• Reduced overtreatment

Sascha Tuchman, MD, of UNC Lineberger leads Alliance A062102, a new study for people living with multiple myeloma. This trial is looking at whether a daily pill called iberdomide can help keep myeloma under control longer compared with the usual approach of simply monitoring the disease after initial treatment.
What the study is trying to learn:
➡️ Can iberdomide help people stay in remission longer?
➡️ Could it improve overall survival?
➡️ How does it compare to regular follow up without additional treatment?
Why this matters:
