Congratulations to Paula Reichel on the publication of her review, "Dietary Antigen Interaction with Intestinal Epithelial Cells," in Current Allergy and Asthma Reports!

In this timely piece, Paula — alongside co-authors Somdutta Chakraborty, Hock Tay, and Simon Hogan reviews the mechanisms by which dietary antigens cross the intestinal epithelium, and how these processes shape oral tolerance versus food sensitization. From GAPs and SAPs to receptor-mediated transport and leukotriene signaling, the review offers a comprehensive look at a rapidly evolving area of food allergy research.

Study Reveals Key Molecular Differences Between PPI-Responsive and PPI-Unresponsive Eosinophilic Esophagitis
New research from our team reveals that pediatric patients with eosinophilic esophagitis (EoE) have distinct molecular signatures that may predict treatment response to proton pump inhibitors (PPIs).

Through comprehensive RNA-sequencing of esophageal biopsies, we discovered that while PPI-responsive (PPI-R) and PPI-unresponsive (PPI-UR) EoE patients appear clinically identical at diagnosis, they exhibit unique transcriptional profiles. PPI-R EoE shows enrichment in interferon signaling and zinc ion regulation pathways, whereas PPI-UR EoE displays heightened type 2 inflammation, enhanced B cell and neutrophil signatures, and—most notably—a sustained esophageal epithelial proliferative response driven by cell cycle and division genes.
Importantly, we identified a proinflammatory SFRP1+ esophageal suprabasal cell population that persists in PPI-UR EoE even after treatment, suggesting a STAT3-dependent mechanism that may explain PPI resistance.

These findings represent a crucial step toward precision medicine in EoE management, identifying potential biomarkers that could guide initial therapy selection and prevent disease progression during ineffective treatment periods.

Dr Hogan and Lukacs recently published a review article in Nature Review Immunology discussing how different routes of exposure to food antigens can contribute to allergic sensitization and describe how mast cells ultimately drive the allergic reaction to these food allergens.

Congratulations to Jazib Uddin, the Hogan Lab and our collaborators, O’Roirdan, Lukacs, Denson, Haberman, O’Dwyer, Divanovic, Weaver, Nusrat and Parkos Labs on the publication of the article in JCI Insight describing a role for STARD7 in intestinal epithelial mitochondrial function and barrier integrity and susceptibility to IBD.
