DoE-derived continuous and robust process for manufacturing of pharmaceutical-grade wide-range LNPs for RNA-vaccine/drug delivery

Lipid nanoparticle (LNP) technology, crucial for delivering RNA-products and drugs, faces a challenge in continuous pharmaceutical-grade manufacturing with a specific particle size range. Our unique platform addresses this gap, producing LNPs ranging from 60 to 180 nm, meeting regulatory standards for polydispersity index, sterility, dose uniformity, and bio-functionality. Employing design of experiment (DoE) methodology, we identified critical parameters, ensuring global applicability. Cross-point validation within the DoE response map confirmed the platform's robustness, consistently producing specific size LNPs (± 10 nm). Successfully scaled to production, our technology, validated through R&D, pilot, and production batches, demonstrates equivalent biological responses for a candidate SARS-CoV-2 mRNA-vaccine. This collective data establishes the platform's robustness and bio-uniformity, vital for global RNA-vaccine/drug formulation.
